Celcuity Inc. is a clinical-stage biotechnology company focused on targeted cancer treatments for patients with solid tumors. Its work centers on understanding and inhibiting signaling pathways that drive tumor growth, with a particular emphasis on hormone receptor-positive, HER2-negative breast cancer, one of the most common breast cancer subtypes.
The company’s lead drug candidate is gedatolisib, an investigational therapy designed to inhibit the PI3K/AKT/mTOR, or PAM, pathway. This pathway is frequently implicated in cancer cell growth, survival, and resistance to endocrine therapy. Gedatolisib is differentiated by its approach to pathway inhibition: it targets all class I PI3K isoforms as well as both mTORC1 and mTORC2, creating a broader mechanism than therapies aimed at a single component of the pathway.
Celcuity is evaluating gedatolisib in several clinical programs. Its Phase 3 VIKTORIA-1 trial studies gedatolisib in combination with fulvestrant, with or without palbociclib, in patients with HR-positive, HER2-negative advanced breast cancer after prior endocrine therapy. The trial has completed enrollment and has reported results for patients with PIK3CA wild-type tumors, while its PIK3CA-mutant cohort has also enrolled.
The company is also advancing VIKTORIA-2, another Phase 3 study in endocrine-resistant HR-positive, HER2-negative breast cancer, and CELC-G-201, a Phase 1b/2 trial exploring gedatolisib combinations in metastatic castration-resistant prostate cancer. As a development-stage biotech, Celcuity’s value is closely tied to clinical outcomes, regulatory progress, and the potential commercial role of gedatolisib in oncology treatment.
