ALX Oncology Holdings Inc. is a clinical-stage biotechnology company developing cancer immunotherapies that are designed to help the immune system identify and attack tumour cells. Its research is centred on the CD47 pathway, a mechanism some cancers use to send a “do not eat me” signal to macrophages and other immune cells. By blocking that signal, the company aims to make malignant cells more vulnerable to immune clearance while improving on the tolerability challenges seen with earlier CD47-targeting approaches.
The company’s lead candidate is evorpacept, also known as ALX148, a next-generation CD47 blocker engineered with an inactive Fc region. This design is intended to bind CD47 with high affinity while limiting unwanted destruction of healthy blood cells, a key concern in the class. ALX Oncology evaluates evorpacept in combination with other anti-cancer treatments, including tumour-targeting antibodies, immune checkpoint inhibitors and chemotherapy, because CD47 blockade is generally viewed as a way to enhance existing therapeutic strategies rather than replace them.
As a development-stage company, ALX Oncology does not have a marketed oncology product and its valuation is closely tied to clinical trial progress, safety data, partnership potential and regulatory outcomes. Its pipeline targets both solid tumours and blood cancers, reflecting the broad role CD47 may play in immune evasion. For investors, ALXO represents exposure to a specialised area of immuno-oncology where scientific differentiation, trial execution and competitive positioning against other macrophage-checkpoint programmes are central to the investment case.
