Cogent Biosciences (COGT)

Its lead programme, bezuclastinib, also known as CGT9486, is a selective tyrosine kinase inhibitor designed to inhibit mutations in KIT, including the KIT D816V mutation and other exon 17 alterations. KIT signalling plays an important role in certain diseases, and abnormal activation can drive unchecked cell growth or mast cell proliferation. Cogent is developing this programme for systemic mastocytosis, a serious disorder marked by the accumulation and activation of mast cells, where KIT D816V is a common underlying driver.

The company is also pursuing opportunities in gastrointestinal stromal tumours, or GIST, a cancer type that can depend on oncogenic KIT signalling. Exon 17 mutations are particularly relevant in advanced or resistant disease settings, making selective inhibition a potential strategy for patients whose disease is not adequately controlled by existing treatments.
